Research Activity

Microglia (MG), the resident immune cells of the Central Nervous System (CNS), are involved in mediating either degenerative responses, characterizing neuroinflammatory disorders, or processes of tissue repair, also occurring in the diseased CNS. Following cell damage, MG processes direct their motility exploring the extracellular space to specifically detect cell damage. The activation of MG cells is relevant for the progression of the degeneration, although molecular cascades triggering this activation are far to be completely elucidated. However, decrypting such cascades may be crucial for the development of new therapeutic tools for the treatment of such disorders. At the same time, the identification of cues orchestrating MG cells to acquire protective functions are also important to foster endogenous CNS repair. We have characterized MG cells in the embryonic brain and we found that MG receive and send instructive signals to neural stem cells of the developing cerebral cortex. Such interactions are necessary for the proper development of the brain. We are exploring such signaling cascades, aiming to identify new druggable pathways for MG modulation in the pathological CNS.

We are also exploring mechanisms, acting on neurons during neuroinflammation, that can trigger neuronal derangement. In this framework, we focus our attention on extracellular acidosis which is accompanying neuroinflammatory/neurodegenerative processes. In physiological conditions, the extracellular pH levels are maintained at 7.4 through various H+ transporting mechanisms. However, during inflammation such levels are persistently reduced, causing long-lasting acidosis in large regions of the CNS. Acid Sensing Ion Channels (ASICs) are voltage independent proton gated cation channels expressed by neurons of the CNS. In mammals, four genes encode at least six different ASIC isoforms, although CNS neurons preferentially express Asic1a and ASIC2(both a and b variants) isoforms. Pharmacological inhibition of ASICs in animal models of Multiple Sclerosis as well as in patients elicits some neuroprotective effects, reducing demyelination and axonal damage in animals and restraining rates of whole-brain atrophy in patients. We are developing new small molecules being for the efficient and specific inhibition of ASICs channels.
