NOTE BIOGRAFICHE

Nicola Clementi ha studiato presso la Scuola di Specializzazione in Microbiologia e Virologia Medica dell’Università “Vita-Salute” San Raffaele di Milano dal 2007 al 2010 dopo aver conseguito la Laurea Magistrale in Scienze Biologiche ad indirizzo biotecnologico presso l’università Politecnica delle Marche nel 2006 con una tesi sperimentale su anticorpi monoclonali umani protettivi diretti contro il virus dell’epatite C (HCV). Dopo la Laurea Magistrale ha frequentato attivamente la SOD di Virologia presso l’Azienda Ospedaliera Universitaria di Ancona sotto la guida del Prof. P. E. Varaldo e della Prof. P. Bagnarelli. Durante la Scuola di Specializzazione, sotto la guida dei Prof. M. Clementi e R. Burioni, ha frequentato il reparto di Microbiologia e Virologia dell’Ospedale San Raffaele, in particolare ha frequentato i reparti di batteriologia, virologia, micologia e parassitologia, conseguendo il titolo di Specialista in Microbiologia e Virologia Medica nel 2010 con una tesi sperimentale incentrata sulla dissezione molecolare della risposta immune umorale umana nei confronti di virus influenzali di tipo A. Nel 2008 si è recato come Visiting Scientist presso il Molecular Medical Science Department diretto dal Prof. J. K. Ball del Queens Medical Centre, Nottingham University Hospital NHS, (Nottingham U.K) lavorando su approcci molecolari per la mutagenesi di geni codificanti per glicoproteine virali di superficie.

 

ATTIVITÀ ACCADEMICA

Dal 2011 al 2015 ha ottenuto numerosi e continuativi incarichi come Professore a Contratto presso la Facoltà di Medicina e Chirurgia dell’Università “Vita-Salute” San Raffaele di Milano suddivisi nei corsi di: “Microbiology and Mechanisms of Infectious Diseases” presso International MD program corso di laurea magistrale in Medicina e Chirurgia; Microbiologia Medica presso il Corso di Laurea magistrale in Medicina e Chirurgia; Virologia Molecolare e Microbiologia Diagnostica presso il Corso di Laurea Specialistica in Biotecnologie Mediche Molecolari e Cellulari; Microbiologia nell’ambito del corso di “Patologia Generale, Immunologia e Microbiologia del Corso di Laurea in Biotecnologie Mediche e Farmaceutiche.

Dal 2011 è docente presso la Scuola di Specializzazione in Microbiologia e Virologia della Facoltà di Medicina e Chirurgia dell’Università “Vita-Salute” San Raffaele di Milano.

Dal gennaio 2016 svolge attività didattica come Ricercatore a TD presso L’Università Vita-Salute San Raffaele di Milano.

È stato inoltre correlatore di tesi di studenti afferenti ai corsi di laurea triennali e specialistiche in biotecnologie.

 

ATTIVITÀ DI RICERCA

Dal 2013 supervisiona e organizza il lavoro sperimentale di dottorandi, tesisti di lauree specialistiche o magistrali e tecnici di ricerca.

La sua attività di ricerca verte principalmente sulla dissezione molecolare della risposta anticorpale umana protettiva nei confronti di un ampio spettro di virus (HCV, HIV, influenza, HSV), batteri e miceti patogeni opportunisti di maggior interesse medico attraverso la costruzione di librerie anticorpali fagemidiche, il clonaggio e la caratterizzazione di anticorpi monoclonali umani. Dal 2015 la sua attività di ricerca si sta particolarmente incentrando sullo studio dell’immunità umorale umana diretta contro virus erpetici quali HSV, VZV e HCMV.

In quest’ambito gli aspetti di maggior rilievo della sua attività di ricerca sono:

 

  • Identificazione e clonaggio di anticorpi monoclonali umani diretti contro microrganismi patogeni ipervariabili di interesse clinico
  • Caratterizzazione molecolare degli epitopi protettivi presenti sulle proteine bersaglio di anticorpi monoclonali dotati di potente attività anti-microbica per la messa a punto di nuovi vaccini epitope-based
  • Ingegnerizzazione di anticorpi monoclonali umani in diversi formati anticorpali dotati di diverse proprietà farmacocinetiche
  • Valutazione dell’attività biologica, terapeutica e/o profilattica di anticorpi monoclonali umani in modelli di infezione in vivo

 

Nell’ambito della Virologia Medica si occupa dal gennaio 2016 dello studio delle farmacoresistenze associate a fallimento terapeutico in pazienti infettati da virus dell’epatite C (HCV) e trattati con i Direct-Acting Antivirals (DAA), farmaci recentemente introdotti nel trattamento delle infezioni da HCV. In quest’ambito assieme al Dott. E. Boeri dell’Unità di Farmacoresistenze del Laboratorio di Microbiologia e Virologia dell’Ospedale San Raffaele si occupa della messa a punto di nuovi test molecolari basati sullo studio di sequenza per l’identificazione e l’analisi di virus mutanti associati a resistenza e fallimento terapeutico in pazienti trattati con farmaci inibitori di NS5A e NS5B di HCV.

BIOGRAPHICAL NOTES

Nicola Clementi was Board Certified in Medical Microbiology and Virology by the “Vita-Salute” San Raffaele University, Milan, Italy in 2010. He earned Biological Sciences Master Degree in 2006 at the Università Politecnica delle Marche, Ancona, Italy. In 2008 was Visiting Scientist at Molecular Medical Science Department (Queens Medical Centre, Nottingham University Hospital NHS, Nottingham UK). During this period he worked on tailored mutagenesis of virus envelope glycoproteins and on alanine scanning techniques.

From 2011 to 2016 he held the position of Contract Professor of Microbiology, Mycology and Parasitology, Molecular Virology and Diagnostic Microbiology at the “Vita-Salute” San Raffaele University, Milan (MD school, International MD program, BMMC, BMF). From 2011, he teaches Medical Microbiology at Microbiology and Virology postgrad specialisation course.

Since January 2016, he is Assistant Professor (ricercatore universitario TD) at Vita Salute San Raffaele University of Milan.

 

His main research interests include:

  • Molecular dissection of the human humoral immune response during HIV, HCV, influenza viruses, HSV, bacterial and opportunistic fungal pathogens infections by the selection and the characterization of broadly neutralizing monoclonal antibodies. The molecular characterisation of protective epitopes recognised by human monoclonal antibodies able to target microbial pathogens in order to develop new epitope-based vaccines.
  • Engineering of human monoclonal antibodies in different molecular formats endowed with different pharmacokinetic and/or pharmacodynamics.
  • In vivo characterisation of biological activity of human monoclonal antibodies able to target clinically relevant microbial hypervariable pathogens.

 

From 2016 January, his work focuses also on the study of the emergence of HCV drug resistant virus mutants in patients treated with the so-called DAA (Direct-Acting Antivirals) recently licensed for human therapy. In particular he perform analysis on HCV NS5A and HCV NS5B gene sequences of viral HCV quasispecies found in HCV patients failing the anti-NS5A and/or NS5B based therapy.
